PURPOSE:To obtain the subject oxidase having substrate specificity to NADH and excellent productivity and useful for the quantitative determination of NADH, etc., by cultivating a specific enzyme-producing microbial strain and adding ammonium sulfate to the cultivation product to effect the salting-out of the enzyme. CONSTITUTION:An enzyme-producing strain Streptococcus mutans is cultivated in a medium composed of glucose, casein, etc., under anaerobic condition at 30-40 deg.C and the cultivation product is salted out with ammonium sulfate to obtain the objective oxidase having a molecular weight of 48,000-52,000 and capable of substrate-specifically oxidizing NADH in the presence of oxygen molecule to form water. |
