PURPOSE:To obtain the subject substance having extremely low toxicity, free from sensitogenicity and continuously administrable without causing side effect by culturing APP strain in a state to minimize the peeling of the membrane surface substance, disintegrating and centrifuging the obtained cell and removing side action substances from the filtrate. CONSTITUTION:APP strain is cultured by gentle shaking culture, standing culture or aeration and agitation culture and the culture product is filtered to collect bacterial cells. The cells are sufficiently mixed with water or preferably a proper buffer solution, disintegrated with a French press, etc., using glass beads under chilling and subjected to centrifugal separation to obtain an extract liquid free from cell. The cell residues are removed from the extract by centrifugal separation and side-effect substances are removed from the solution by column chromatography, ultrafiltration, extraction with organic solvent, affinity chromatography with monoclonal antibody or isoelectric precipitation to obtain the objective phylaxis immunity substance against APP infection disease. |
