摘要 PROBLEM TO BE SOLVED: To inspect the mutation of a plurality of base sequences with only once operations in series of the PCR gene amplification, heating, reconjugation and analysis. SOLUTION: This mutation detection process comprises the steps wherein (A) a normal type DNA 15 and a mutation type DNA 17 are mixed and the mixture is subjected to the PCR amplification; (B) fluorescence-labeled oligonucleotides 19a, 19b and 19c that are individually complementary to any of exons 15a, 15b, 15c and are labeled with fluorescent substances F1, F2, and F3 that have individually different fluorescent spectra are prepared; (C) individual oligonucleotides are hybridized to the amplified products to form the homo type conjugation site 21a, 21b, 21c, 23c and the hetero type conjugation sites 23a, 23c; (D) since the hetero type conjugation site has a lower melting point than that of the homo type conjugation site, they are analyzed with the ion-pair chromatography of which the reversed phase column is adjusted to the melting point to detect the fluorescence-labeled oligonucleotide 19a, 19b, 19c that are eluted at the relatively early stage with discrimination thereby detecting the exons causing the mutation.
