PURPOSE:To improve efficiency of enzymatic reaction in producing alpha-tocopherol from non alpha-tocopherol by using an enzyme capable of converting tocopherolmethyl group, by adding a specific metallic ion to the reaction system and making a water-soluble organic solvent exist. CONSTITUTION:In producing alpha-tocopherol by treating non alpha-tocopherol with an enzyme (e.g. chloroplast prepared according to intracellular granule fractionation method from plant or alga) capable of converting tocopherolmethyl group in the presence of a methyl group donor (preferably active methionine) under a weakly alkali condition (preferably pH7.5-8.5), preferably 0.01-1mM one or more metallic ions selected from the group consisting of Mg, Ca, Zn and Sn and preferably 5-30vol% water-soluble organic solvent (preferably methanol, ethanol, acetone, DMF or DMSO) are added to the reaction system.
