The present invention relates to recombinant rabies virus, a recombinant rabies virus vaccine composition including the same, and multiplex RT-PCR used to detect the same. More specifically, the recombinant rabies virus is prepared by replacing the 333rd amino acid in G protein of the rabies virus with a glutamic acid and the 194th amino acid with serine to enhance the effect and stability. The multiplex RT-PCR can be used to accurately identify the outdoor rabies virus. The recombinant rabies virus is highly safe and can be used for oral administration to solve the problems of conventional rabies vaccine. According to the present invention, a method for preparing the recombinant rabies virus can be used to efficiently produce the excellent recombinant rabies virus in host cells, distribute the vaccine through mass-production, and significantly contribute to the prevention thereof. According to the present invention, a recombinant rabies virus detection method or a recombinant rabies virus detection kit can be used to accurately identify and distinguish the infection of the recombinant rabies virus and the outdoor infection of the recombinant rabies virus in order to easily determine whether the vaccine is administered to a wild animal or whether the wild animal is infected and is capable of inducing the rabies infection.
