The present invention provides assays and methods for determining levels of STIM2 activity, thus providing tools for the characterization and study of the regulation of intracellular calcium levels. In specific embodiments, the invention relates to assays for STIM2 activity in a cell comprising measuring Icrac activity; assays for agents that modulate STIM2 activity comprising measuring Icrac activity in response to application of 2-APB in the absence of calcium store depletion; and to assays to screen for an aminoglycoside antibiotic that affect immune cells, the assay comprising measuring Icrac activity after application of the aminoglycoside antibiotic.
