摘要 PROBLEM TO BE SOLVED: To improve accuracy in estimation of an amino acid sequence using de novo sequencing without requiring database, and, in particular, to provide one or a small number of sequences which are correct answers.SOLUTION: An analysis method for an amino acid sequence includes the steps of: subjecting to mass spectrometry a plurality of samples obtained by fragmenting a plurality of enzymes each having a different cleavage site, and executing de novo sequencing to the result obtained to estimate a partial sequence candidate for each of the fragments (S1, S2); extracting a partial sequence candidate containing the terminal of the original amino acid from a large number of the partial sequence candidates by using a phenomenon that a specific amino acid residual group appears on the cleavage site according to the enzyme (S6); further repeating an operation of retrieving and bonding partial sequence candidates having overlapping portions among the partial sequence candidates other than those containing the terminal (S7, S8). When a sequence is wrongly estimated by the de novo sequencing, it is hardly possible that a large number of sequence candidates can be bonded to each other without contradiction. Therefore, the analysis method further includes the steps of: adding the sequence candidate containing the terminal (S9); and increasing the possibility that the finally produced amino acid sequence is a correct answer (S10, S11).
