OPTICAL MICROSCOPY USING PHOTO-CONVERTIBLE OPTICAL LANDMARK

PROBLEM TO BE SOLVED: To provide optical microscopy using photo-convertible optical landmarks.

SOLUTION: First activation radiation is supplied to a sample 601 including photo-convertible optical landmarks ('PTOLs') to activate a first subset of the PTOLs in the sample. First excitation radiation is supplied to the first subset of the PTOLs in the sample to excite at least some activated PTOLs, and radiation to be emitted from the activated and excited PTOLs in the first subset of the PTOLs is detected by an optical element 606 for imaging. The first activation radiation is controlled so that an average volume per activated PTOL in the first subset becomes almost equal to or larger than a diffraction limit resolution volume ('DLRV') of the optical element 606 for imaging.
