Disclosed are insect cell lines in vitro. In some embodiments, cells are derived from primary cell cultures from eggs of the black witch moth Ascalapha odorata. Also disclosed are methods of generating the cell lines, and methods of using the cell lines for expressing polypeptides encoded by exogenous nucleic acids. In some embodiments, A. odorata cells in vitro can produce a polypeptide encoded by an exogenous nucleic acid at a level greater than that of insect cell lines Sf9 or BTI-TN-5B1-4. In various configurations, an A. odorata cell line can have a doubling time less than that of BTI-TN-5B1-4 cells, and can grow in a serum-free medium. In some configurations, an A. odorata cell line can be free of an alphanodavirus. In some embodiments, A. odorata cells in vitro can secrete recombinant polypeptides in glycosylated form. |
