<P>PROBLEM TO BE SOLVED: To provide an FRET measuring technology capable of accurately obtaining a dissociation constant K<sb>d</sb>independently of size of the dissociation constant K<sb>d</sb>. <P>SOLUTION: In an FRET measuring method, FRET that an energy transfers from a first molecule to a second molecule is measured by applying a laser beam to a measurement sample which has a ligand-bound receptor and is provided with the first molecule and the second molecule. The FRET measuring method has steps of: applying a laser beam to the measurement sample; measuring fluorescence produced by the measurement sample; calculating a fluorescence lifetime of the first molecule; calculating a binding ratio; setting up binding conditions of the measurement sample; and calculating a dissociation constant. In the step of calculating the dissociation constant, a least-square method is used to obtain the dissociation constant through fitting of function with setting a total concentration of the receptor in the measurement sample and the dissociation constant as a variable into the binding ratio calculated in the step of calculating the binding ratio. <P>COPYRIGHT: (C)2012,JPO&INPIT |
