PROBLEM TO BE SOLVED: To provide a method of manufacturing an optical waveguide and opto-electric composite substrate capable of avoiding a case where a remaining core or a foreign substance occurring near a patterned core layer directly comes into contact with the patterned core layer and suppressing degradation of the optical transmission loss, and to provide an optical waveguide and opto-electric composite substrate obtained by the method. SOLUTION: This method of manufacturing the optical waveguide and opto-electric composite substrate includes a first process of forming a substantially rectangular member consisting of a lower clad layer of a substantially rectangular shape on a base material or electric wiring board and a core layer of a substantially rectangular shape mounted on the top surface of the lower clad layer, and a second process of forming an upper clad layer for covering the top surface and side surface of the core layer and the side surface of the lower clad layer.
