PROBLEM TO BE SOLVED: To provide a spacer for a culturing container without using a special apparatus, only by applying it to a generally used culturing container such as a multi-well plate with a lid body, capable of observing the effects of an objective substance contained in a gas phase to a living body sample by real time, evaluating in a good reliability and also without being limited by the kind of objective substances, a culturing apparatus including the spacer, and a method of observation by using the culturing apparatus. SOLUTION: This spacer 10A for the culturing container is provided by being attached between a culturing container body 21 of the culturing apparatus and a lid body 22 being covered on the culturing container body 21, and while separating the culturing container body 21 and lid body 22, air-tightly connecting them. By the culturing apparatus equipped with the spacer 10A, while culturing the living body sample, it is possible to observe the effects of the substance contained in the gas phase in the culturing apparatus to the living body sample in real time.
