1,237,852. Determination of lactic acid. SMITH KUNE & FRENCH LABORATORIES. 29 Aug., 1969 [6 Sept., 1968], No. 43232/69. Heading B1X. A reagent for determining lactic acid comprises a solid, water soluble substantially anhydrous mixture of (a), lactate dehydrogenase (LDH) and glutamate-pyruvate transaminase (GPT); (b), NAD; (c), as pyruvate trapping agent, glutamic acid or its alkali metal salt; and (d), a buffer to maintain the pH between 7.5 and 11.0; and preferably also (e), a stabilizer for the enzymes selected from mucilagenous gum, hydroxyalkylamines, EDTA and its salts, a source of sulphate anion, and mixtures thereof; and (f), a stabilizing-bulking agent selected from mannitol, sorbitol, lactose and polyvinyl alcohol. Other stabilizers are mentioned. A solution of the enzymes (a) and stabilizer (e) may be lyophilized and the residue mixed with the other dry components. The reagent may be packed in unit amounts in for example gelatin capsules in packages containing an alumina pellet as desiccant.
|
