PROBLEM TO BE SOLVED: To clarify a structure and functionals of a homingendonuclease encoded by COB gene group I intron of C. smithii and provide the homingendonuclease as a DNA-cleaving enzyme.

SOLUTION: An isolated or refined polypeptide composed of a part of a protein encoded by a translation region in COB gene group I intron of C. smithii and composed of a specific amino acid sequence or a shortened amino acid sequence in which 1-37 arbitrary amino acids are lost from the N end of the above amino acid sequence or its salt.
