<P>PROBLEM TO BE SOLVED: To provide an amino acid sequence of an SM synthase and a base sequence encoding the same, and new uses of the same. <P>SOLUTION: This human cDNA (SMS-1) corresponding to the SM synthase activity is isolated by an expression cloning method by using a leukemic cell mutant (WR19L/Fas (-)) of a mouse, lacking the SM synthesis. The WR19L/Fas-SM(-) cells do not express the SM, but the SMS-1 cDNA-expression cells express the SM in a same degree to that of the WR19L/Fas-SM(+) cells, and also since a ceramide and phosphatidylcholine are required for the expression of the SM, it encodes the SM synthase. Also, the SM synthase encoded by the SM-1 cDNA is essential for the cell proliferation. <P>COPYRIGHT: (C)2005,JPO&NCIPI |
