PURPOSE: A method for purifying an acidic polysaccharide derived from a microorganism is provided, thereby effectively removing the microorganism with improved homogenization condition using the reduced amount of isopropanol, so that recover rate of the acidic polysaccharide can be increased. CONSTITUTION: The method for purifying an acidic polysaccharide derived from a microorganism comprises the steps of: culturing an acidic polysaccharide producing microorganism in TSB medium at 30 deg. C for 1 day; separating single colony grown and inoculating on TSB medium containing rifampicin and culturing it at 30 deg. C and 250 rpm for 22 hours; inoculating the cultured medium into the restriction medium for synthesis of polysaccharide, diluting the medium with distilled water, and homogenizing the mixture at 55 to 60 deg. C and 5,000 rpm for 30 minutes; centrifuging the homogenized mixture at 8,000 rpm and 25 deg. C for 30 minutes to remove microorganism pellet; treating the supernatant with NaOH; adding acetic acid into the NaOH treated supernatant to neutralize it; and adding the same amount of isopropanol into the neutralized mixed solution to recover clot type polysaccharide, wherein the acidic polysaccharide producing microorganism is Zoogloea ramigera 115SLR or Xanthomonas campestris.
