PURPOSE: A metal chelate resin, its preparation method and a method for purifying proteins and a method for fixing proteins by using the metal chelate resin are provided, to improve the adsorption and stability to the materials to be separated by using the chelate resin. CONSTITUTION: The metal chelate resin is represented by (CH2COOH)2-N-CH(COOH)-CH2CH2-CO-NH-CH2(CH2S-R1-R2)-CO-NH-CH2(COOH), wherein R1 is a linker which activates a polymer matrix for chromatography and combines with -SH group; and R2 is a polymer matrix for chromatography. Preferably the polymer matrix for chromatography is at least one selected from the group consisting of a crosslinked agarose, a crosslinked cellulose, a crosslinked dextran, a crosslinked methacrylamide, proteins, peptides, lipids, liposome, nucleic acids, antibodies, enzymes, polyethylene, polysulfone, gold, hormones, fluorescent materials, chromogenic dyes, silica and glass. Preferably the metal chelate resin is combined with a transition metal ion. The preparation method comprises the steps of activating a polymer matrix; washing the activated one; reacting the polymer matrix with a peptide having -SH group at a pH of 6.0-8.5; washing the obtained one; and combining acetic acid to an amine group at a pH of 9-12. Preferably the peptide having -SH group is N,N-di-carboxymethyl glutathione.
