A microfluidic device (100) for achieving protein separation based on charge and molecular weight or size is provided and includes: (1) a first substrate (110) having a raised structure protruding from a first face (112) of the first substrate (110) such that the raised structure partially defines a microfluidic cavity (130); (2) a second substrate (120) having a raised section protruding from a first face of the second section. The microfluidic device (100) also includes: (3) sample inlet (115) and outlet (116) ports formed in the second substrate (120); (4) buffer and gel input and output ports (117) formed in the second substrate (120); and (5) a plurality of ports (140) formed in the second substrate (120) for sample selection and sample output.
