<P>PROBLEM TO BE SOLVED: To provide a method for efficiently obtaining a variant antibody having improved integrity with a material having antigenicity. <P>SOLUTION: The method for obtaining the variant antibody involves using a variant Fv library which is a domain included in one CDR of an antibody or present at the vicinity thereof, designed so that k (k is an integer satisfying the relation of 2≤k≤20) kinds of amino acid mutation may be introduced to m (m is an integer satisfying the relation of 2≤m≤n-1) residues of amino acids selected from n (n is an integer satisfying the relation of 3≤n) residues of amino acids, and provides<SB>n</SB>C<SB>m</SB>×k<SP>m</SP>kinds of limited Fv repertories as amino acid sequences. Preferably, the variant Fv library in which k is 20, which is designed so that NNK (N is A, G, C or T; and K is G or T) mutation may be introduced as a nucleotide residue corresponding to the amino acid residue to which the mutation is introduced, and which provides the limited Fv gene repertories of<SB>n</SB>C<SB>m</SB>×32<SP>m</SP>kinds as the base sequences, is used. <P>COPYRIGHT: (C)2004,JPO |
