PURPOSE: A method for producing a cell-free protein and a method for producing a protein required to be post-translational modification using the same method are provided, thereby several proteins for medical treatment can be cheaply produced. CONSTITUTION: The coupled transcription/translation method for producing the cell-free protein comprises the steps of: adding DNA encoding protein to be produced into a cell extract; adding nucleic acid and energy source into the cell extract; and adding intracellular tools associated with the post-translational modification containing endoplasmic reticulum/Golgi body, endoplasmic reticulum/Golgi body/cell membrane, or other components into the cell extract. in which the protein is EPO(erythropoietin), the post-translational modification is the addition of sugar chain.
