摘要 PURPOSE: The osmotic stress resistant potatoes transformed by a recombinant vector containing the GPD gene isolated from Pleurotus sajor-caju is provided for improving the osmotic stress resistance of potatoes, so that the potatoes can be grown in soil slightly containing salt. CONSTITUTION: The osmotic stress resistant potatoes transformed by a recombinant vector containing glycerol-3-phosphate dehydrogenase(GPD) gene as an osmotic stress related gene is obtained by the following steps of: incubating Pleurotus sajor-caju ASI 2070 in an MCM medium containing 20g of peptone, 2g of yeast extract, 2g of K2HPO4, 1.0g of KH2PO4, 0.46g of MgSO47H2O, 0.5g of DW1L at 25deg.C for 15 days; filtering with mira cloth; separating total RNA and DNA of Pleurotus sajor-caju ASI 2070 by using a Graham method and a Sambrook method, respectively; preparing pBI vector containing the GPD gene by using restriction enzymes; infecting Agrobacterium with the pBI vector; and transforming potatoes with the pBI infected Agrobacterium.
