PROBLEM TO BE SOLVED: To obtain a new trehalose phosphorylase excellent in temperature stability and useful for production, etc., of glucosyl transfer sugar-containing saccharide from a microorganism belonging to the genus Thermoanaerobium. SOLUTION: This trehalose phosphorylase can be obtained from a microorganism belonging to the genus Thermoanaerobium, e.g. Thermoanaerobium brockii, having action of decomposing trehalose in the presence of an inorganic phosphoric acid (salt) and producing D-glucose and β-D-glucose-1-phosphoric acid (salt) and has the following physicochemical properties: Molecular weight, 88,000±5,000 dalton (by SDS electrophoresis); optimum temperature, 7.0-7.5 when reacted at pH 7.0 for 30 min; temperature stability, stable up to nearly 60 deg.C when retained under conditions of pH 7.0 for 1 hr; pH stability, 6.0-9.0 when retained at 4 deg.C for 24 hr. Furthermore, the phoshorylase contains an amino acid sequence, etc., of formula I as a partial amino acid sequence and contains an amino acid sequence of formula II and is obtained by culturing a microorganism having trehalose phosphorylase-producing ability. |
