PURPOSE: To obtain squalene which is a starting substance of squalane useful as an oily starting substance for cosmetics by culturing a protozoa in Euglena in a nutrient culture medium under aerobic conditions and collecting the product from the culture mixture. CONSTITUTION: A protozoa in Euglena such as Euglena gracilis SM-ZK strain is cultured aerobically under shaking at 100 times/min at 25 deg.C for 5 days. Then, the culture mixture is charged in a flask containing a culture medium to which further carbon source and vitamins are added and the flask is tightly closed with a paraffin film or the like, and the culture is continued under anaerobic conditions for 1-3 days. After completion of the culture, the culture mixture is centrifuged to collect the cell bodies, which are dispersed in a chloroform: methanol=2:1 mixture to effect shaking extraction. Then, the cell bodies are removed and the solvent is distilled off the extract to give squalenes which gives squalane as an oily starting substance for cosmetics. |
