<p>PURPOSE:To produce the subjcet RNA used for controlling cucumber mosaic viruses, etc., by recombining a satellite RNA containing a specific base sequence with a complememtary cDNA, introducing the produced plasmid into Escherichia coli, culturing the formed transformant and subsequently treating the cultured transformants witn an enzyme. CONSTITUTION:The cotyledons of a tomato are inoculated with the ground solution of tomato leaves infected with the weak toxic virus of cucumber mosaic virus(CMV) containing a satellite RNA, and the virus is multiplied for approximately one to four weeks. The infected tomato leaves are collected, frozen, pulverized and subsequently ground in a buffer solution. The ground solution containing the virus particles is centrifuged, and the upper layer is collected and subsequently mixed with a powdery polyethylene glycol to deposit the virus particles, which are centrifuged to recover the virus particles and in corporated with a buffer solution containing a surfactant to be dissolved. The recovered virus supernatant is collected, mixed with phenol to decompose the outer coating proteins of the virus particles, and subsequently centrifuged. The supernatant containing RNA is separated and subsequently centrifuged by a scurose density gradient method. The satellite RNA of the formula is isolated by an electrophoresis method.</p> |
