<p>PURPOSE:To improve the production of a viscous polysaccharide useful as a raw material for drugs, foods, etc., by culturing a plant tissue as a callus thereof in a liquid medium. CONSTITUTION:A part of the tissue, e.g. leaf, cotyledon or seed, of the Malvaceae plant such as Hibiscus or Abelmoschus is cultured in a basic medium containing 1-10muM of auxin and 0.01-1muM of cytokinin at 25-30 deg.C to induce the callus thereof. The callus is subjected to a subculture in an agar medium containing 1-10muM of auxin and 0.01-5muM of cytokinin for 2-4 months and the multiplied callus is subjected to a shaking culture in a liquid medium containing the same concentrations of the auxin and cytokinin in a dark atmosphere to produce the objective viscous polysaccharide.</p> |
