Fixation and transport of muscle biopsy samples - in 3,3-di:methoxy-propane, preventing loss of enzymatic activity

Fixation and transport of human and animal tissue material for immune- and enzyme- histological test reactions, with both formalin-compatible and -incompatible antibodies (Ab), comprises introducing the material (biopsy) into a liq. contg. 2,2-dimethoxypropane (DMP), present in the transport container. Before the est reaction is carried out, the sample is removed from the MDF and reactivated for 10-15 min (depending on sample size) in an aq. soln. of pH 7.4. Pref. the transport container is a plastic (teflon or polypropylene) or glass tube, contg. about 10ml DMP. USE/ADVANTAGE - The process is applied to muscle biopsy samples, for diagnosis of neuromuscular disease. It allows the sample to be transported (for a week or more) without loss of enzymatic activity or structural integrity. Conventional dewatering treatments are not required.
