PURPOSE:To make the title sensor excellent in heat resistance and solvent resistance and to enable the use of the sensor even in a state of high temperature of a specific temperature or above by a method wherein an enzyme fixing film prepared by fixing an enzyme deriving from a thermophile by an electrolytic polymerization method is provided on a conductive base. CONSTITUTION:In a sensor, a conductive film 11 made up of an iridium oxide is formed on a sapphire board 10. One end side of the conductive film 11 is connected electrically to a lead wire 12 through a conductive bonded film 13 and also covered with an insulative bonded film 14. Besides, an enzyme fixing film 15 is laminated on the other end side of the conductive film 11. The enzyme fixing film 15 is prepared by fixing on an electrolytic polymerization layer of pyrrole an enzyme, e.g. glucokinase, deriving from a thermophile which can grow without losing activity even at a temperature of 40 deg.C or above. When the sensor is immersed in a solution, together with a reference electrode and an opposite electrode, and a change in a current is measured, the concentration of a hydrogen ion changing with the advance of an enzyme reaction is detected as a change in an electromotive force since the iridium oxide is used as a conductive base, and thereby the concentration of a substrate can be measured. |
