<p>NEW MATERIAL:An anti-human pancreas lipase monoclonal antibody having the following properties. Action, recognizing human pancreas lipase; antigen activity after antigen-antibody reaction, free from inhibition of human pancreas lipase activity even after reaction with human pancreas lipase acting as an antigen; kind, IgG1; competitivity, non-competitive to the reaction with labeled human pancreas lipase and non-labeled human pancreas lipase. USE:Reagent for quantitative determination of human pancreas lipase. PREPARATION:The objective monoclonal antibody can be prepared e.g., by (1) emulsifying human pancreas lipase with Freund's complete adjuvant, (2) immunizing BALB/C mouse with the emulsion (3) collecting the spleen cell after the final immunization, (4) fusing the cell with mouse myeloma cell and culturing the fused cell in HAT medium to obtain a hybridoma, (5) screening and then cloning the hybridoma by limiting dilution analysis and (6) culturing the cloned product.</p> |
