PURPOSE:To efficiently refold an urokinase precursor-like protein, by bringing the above-mentioned protein into contact with an alkaline aqueous solution in the presence of a protein denaturing agent, adding an acidic substance in the presence of a saccharide and reducing the pH of the solution. CONSTITUTION:An urokinase precursor-like protein produced as an insoluble paraprotein in a host cell is brought into contact with an alkaline aqueous solution in the presence of a protein denaturing agent. An acidic substance is then added to reduce the pH of the aqueous solution to refold the urokinase precursor-like protein. After the contact with the afore-mentioned alkaline aqueous solution, the pH is preferably reduced in the presence of a saccharide and a sulfhydryl compound. An aqueous solution of monoethanolamine, diethanolamine, etc., is preferred as the alkaline aqueous solution. Guanidine hydrochloride, urea, etc., are cited as the protein denaturing agent.
