Method of producing and purifying a novel human tissue plasminogen activator composition. The tPA composition is produced in myeloma cells, preferably in the presence of epsilon amino caproic acid. It may be purified from the culture media by sequential fractionation on controlled pore-size glass and on affinity chromatography media. It comprises a mixture of proteins which is more homogeneous than the tPA composition produced in CHO cells. It is predominantly single chain material. It comprises a mixture of two chains, a major amount of a sequence beginning with Ser + 1 (mature form), and a minor amount of a sequence beginning with Gly-3. The disclosure enables large scale production of an active human tPA composition very similar to natural endogenous human tPA. |
