PURPOSE:An an aqueous extract of acylase containing protease is circulatively passed through an ultrafilatration membrane and the resultant concentrate is kept at a specific pH whereby the aqueous solution of acylase is kept stable for a long period of time. CONSTITUTION:A filamentous fungus in Aspergillus such as Aspergillus ochraceus IFO 4346 is inoculated in a medium and cultured at 20-40 deg.C for 2-7 days to prepare a culture medium for acylase production. Then, water is added to the medium to effect extraction at 5-30 deg.C for 2-48hr to obtain an aqueous acylase extract containing 100-200 units/ml of acylase and 100-500 units/ml of alkali protease. The aqueous extract is circulatively passed through an ultrafiltration membrane which can fractionate molecules larger than protease and smaller than acylase, at 5 deg.C to room temperature and 1-2kg/cm<2> pressure to obtain a concentrate containing 100-500 units/ml of acylase and 100-500 units/ml of alkali protease. The concentrate is stored with pH kept at 6-7.
