<p>4-(2-Diacetylamino 3,5-dibromo phenyl) methyl) amino trans-cyclohexanol; for use as expectorant; is made by (a) brominating o-toluidine under mild conditions to obtain the 2,4-dibromo cpd. and stronger bromination to give N,N-diacetyl 2,4-dibromo 6-bromo-methyl aniline; (b) aminating with hexamethylene tetramine to give N,N-diacetyl 2,4-dibromo 6-aminomethyl aniline; (c) reacting with cis-4-iodo cyclohexanol in apolar solvent e.g. CCl4 or benzene, in presence of an organic base, e.g. triethylamine to give 4-(2-diacetylamino 3,5-dibromo phenyl) methyl)amino trans-cyclohexanol; and (d) de-acetylating by heating with aq. acid to obtain the required prod. as the hydrochloride.
