PROBLEM TO BE SOLVED: To provide a method for preparing a transgenic plant using an artificial chromosome. SOLUTION: The method for preparing the transgenic plant comprises the steps of transducing a DNA fragment containing a selectable marker into a cell, growing the cell under a selective condition so as to prepare the cell that has incorporated the DNA fragment into its genomic DNA, selecting a cell having a mini chromosome of 10Mb to 50Mb containing a new centromere, the selectable marker and a true chromatin, isolating the mini chromosome, transducing the mini chromosome into a cell of a plant, and culturing the cell of the plant under the condition whereby the transgenic plant is prepared.
