PURPOSE:To obtain a liquid chromatographic analyzing method so as to raise a separation degree, by setting the concentration difference between developing liquids within the range to neglect the shrinkage of packing and varying pH value at the same time so as to increase the elution speed. CONSTITUTION:In a liquid chromatographic analyzing method used a center injection method, a dead space is not formed and accuracy of a quantitative analysis of each component is improved remarkably by sending a developing liquid successively having the concentration not causing the shrinkage of packing which is filled in a separation column and also, increasing pH value successively so as to promote the elution speed of the component. For example, at the time of separating twenty kinds of amino acid, 0.2N, 3pH developing liquid for conditioning is sent to the separation column at first and hereafter, 0.2N, 10pH developing liquid is sent simultaneously with sample injection. |
