Lysergic acids, viz. d-lysergic acid (I) and d-isolysergic acid (II), are prepd. by (a) hydrolysing ergot alkaloids with alkali metal hydroxide and dithionite in aq. soln. at elevated temp. in an inert atmos; (b) concentrating and cooling the hydrolysis mixt.; (c) separating a resinous layer contg. the alkali metal lysergates; and (d) purifying this layer by chromatography on a porous gel. (I) and (II) are intermediates for peptide alkaloids (ergocornine, ergocryptine, ergonin and ergopeptin), amide derivs. ( e.g. ergometrine and LSDe, and ergoline and ergolene derivs. Crude starting materials can be used, e.g. Crude alkaloids or alkaloid mixts. from saprophytic cultures or mother liquors remaining after allakoid sepn. Yields of 81-86.5% can be obtd. |
