A colorimetric sensor for specific detection is provided to improve observation convenience of a target material by changing color of the colorimetric sensor from blue into red and conferring fluorescence to the color-changed part, so that the colorimetric sensor replaces the diagnosis system by using gold particle and fluorescence labeling manners. A colorimetric sensor for specific detection is prepared by (1) dissolving diacetylene monomer of which outer exposed group is substituted by a nonionic group such as hydroxy group and diacetylene monomer having a specific functional group selected from protein, antibody, antigen, DNA, RNA, enzyme, biotin, maleimide and lipid in organic solvent, (2) dipping a support in the mixed solution, (3) pulling out the support and drying the solvent of the support to perform self-assembly, and (4) polymerizing the self-assembled diacetylene monomer to the support through UV(ultraviolet) irradiation.
