EXPRESSION VECTOR FOR CONDITIONAL CONTROL OF EXPRESSION OF RNA POLYMERASE II LARGE SUBUNIT

PROBLEM TO BE SOLVED: To obtain a new expression vector comprising, e.g. a sequence encoding a large subunit ofα-amanitin resistive RNA polymerase II, a sequence allowed for a conditional control of expression or a sequence for duplication, and useful for a stable expression of the above enzyme for an eucaryote. SOLUTION: This new vector contained (A) a nucleotide sequence encoding a gene of a large subunit of RNA polymerase II resistive toα-amanitin or its derivative, (B) a nucleotide sequence enabling a conditional control of an expression of the large subunit, (C) a nucleotide sequence for selection of the vector in cells e.g. eucaryotes or prokaryoes, (D)) nucleotide sequence for duplication of the vector in cells e.g. eucaryotic or prokaryotic cells, and (E) a polyadenylated site (PAA), and it is allowed to stably express a RNA polymerase II large subunit into an eucaryote and ptocaryote in a controllable way.
