The present invention relates to a composition for promoting the biosynthesis of oleanane-based ginsenoside (Ginsenoside Ro). More specifically, the present invention relates to a composition for promoting the biosynthesis of oleanane-based ginsenoside, which contains CYP716A52v2 protein involved in the oleanane-based ginsenoside and a CYP716A52v2 gene encoding the protein. According to the present invention, the composition of the present invention can be useful as a method for increasing the biosynthesis of oleanane-based saponin in ginseng or for mass-producing the oleanane-based ginsenoside.
