PROBLEM TO BE SOLVED: To provide a composition and a method for influencing a regulatory site in the FX zymogen→protease transition pathway thereby driving production of a more "zymogen-like" FXa species, in Factor X/Factor Xa zymogen/protease which modulates hemostasis.SOLUTION: Nucleic acid cloned into an expression vector encodes variant zymogen/protease, and optionally encodes an intracellular PACE/furin cleavage site. A host cell expresses the zymogen/protease variant. A method for treatment of a hemostasis related disorder comprises the step of administration of a therapeutically effective amount of zymogen/protease in a biologically acceptable carrier.
