PROBLEM TO BE SOLVED: To provide an isolated antibody or antigen-binding portion thereof that specifically binds to human CD40 and functions as a CD40 agonist.

SOLUTION: The antibody or antigen-binding portion thereof comprises an isolated nucleic acid molecule including a nucleic acid sequence that encodes a heavy chain or an antigen-binding portion thereof or the light chain or an antigen-binding portion thereof of a human anti-CD40 antibody, wherein nucleic acids, that encode fragments of the heavy chains and the light chains from a hybridoma cell that produce anti CD40 monoclonal antibodies derived from mice immunized with human CD40, are subjected to cloning and sequencing.
