Disclosed are a primer set, a method and a kit for the method, which can achieve the detection of a cancer cell induced by an HPV or the determination concerning whether or not a tissue of interest is in a later stage of severe dysplasia with high accuracy and in a simple manner. The primer set comprises: a first primer which is capable of hybridizing with a nucleic acid comprising a nucleotide sequence that has the substitution of a cytosine residue located in a region outside of a CpG site in a nucleotide sequence containing the CpG site in an L1 or L2 region of an HPV by another base; and a second primer which is capable of hybridizing with a nucleic acid comprising a nucleotide sequence that has the substitution of a cytosine residue located in a nucleotide sequence containing a CpG site in an LCR or an E6 region of an HPV by another base.
