PROBLEM TO BE SOLVED: To provide a manufacturing method of a filler for separation treatment which reduces the influence of a metal in a polymer type filler to constitute the hindrance, in analyzing a compound which exists in liquid in order to accurately purify, concentrate, analyze and sample the extreme trace amount of the compound that is amenable to the influence of a metal, to provide a filler for separation treatment, to provide a cartridge for extracting the solid phase, to provide a column for liquid chromatography and to provide a separation method of a substance to be separated. SOLUTION: The manufacturing method of the filler for the separation treatment comprises bringing the polymer type filler into contact with an organic solvent to swell the polymer type filler and then bringing the same into contact with an acid such as nitric acid and hydrochloric acid.
