It is intended to provide a method of specifically measuring glycolated beta-chain N-end of glycolated hemoglobin by using an enzyme without resort to any separation procedure, and a measurement reagent kit. A protease capable of cutting out a glycolated amino acid and/or a glycolated peptide form glycolated beta-chain N-end of glycolated hemoglobin or its fragment substantially without cutting out any glycolated amino acid or glycolated peptide from glycolated alpha-chain N-end is screened. By using the protease obtained by this screening method, a method and a measurement reagent kit by which the glycolated beta-chain N-end of glycolated hemoglobin can be specifically measured without resort to any separation procedure can be provided. Thus, it becomes possible to specifically measure the glycolated beta-chain N-end of glycolated hemoglobin without resort to any separation procedure.
