PROBLEM TO BE SOLVED: To provide a recombinant vector for the gene therapy of insulin- dependent diabetes and composition for treating the same. SOLUTION: K14 promoter, a human insulin gene and a vector, in which K14 promoter and β-galactosidase gene were cloned, were prepared and were administered into the mouse skin tissue together with a helper vector expressing a Drosophila melanogaster P-transposase by the liposome method and the pressure injection method, so that β-galactosidase gene was expressed from 24 h to 20 weeks, and was inserted into the mouse chromosome by the transposase to observe that β-galactosidase gene was expressed in the keratinocytes layer assuring the enhancing and tissue specificity of the promoter. Although there are almost no β-cells forming insulin in the pancreas of a diabetic mouse, insulin formed from keratinocytes by the expression of the insulin vector lowered the blood glucose level, so that it was found that the insulin vector for the gene therapy has an excellent effect that permits treating diabetes. |
