PROBLEM TO BE SOLVED: To obtain a superoxide dismutase I derived from the mycelium or fruit body of Lentinus edodes having specific enzymatic characteristics, capable of eliminating harmful active oxygen by the enzymatic characteristics, excellent in safety and useful as a treating agent, a cosmetic or the like. SOLUTION: This superoxide dismutase is contained in the mycelium and fruit body of Lentinus edodes and has the following enzymatic characteristics: pH stability: stable at 4-10; heat stability: up to 37 deg.C, 90% activity or more is held, and at 60 deg.C about 30% activity is held; molecular weight: 47000 by gel filtration and 23500±500 by SDS-PAGE; isoelectric point: pH 4.84±0.1; the absorbing spectrum of protein: the maximum absorbing peak exists in the range from 278 nm to 479 nm; contained metal: Mn2+ (the visible absorption of protein of (square)max=479 nm); and the durability of H2O2 inhibition: 80% residual activity or more at 30 min of contact time. Further, the superoxide dismutase I is obtained by aerobically culturing Lentinus edodes which generates superoxide dismutase I, and extracting the obtained mycelia and fruit bodies. |
