PROBLEM TO BE SOLVED: To obtain the subject novel compound that is useful as a metalloproteinase inhibitory substance. SOLUTION: This compound is represented by formula I ((m) is 1-4; (n) and (p) are each 0-4; X is O or the like; R1 is H or the like; R2 -R4 are each H or a 1-6C alkyl; R5 is a halogen or the like; R6 -R8 are each H, a 1-6C alkyl or the like; R9 is SO3 H or the like), typically 2-[(4-methoxyphenyl)-sulfonyl]-9-[(3- morpholin-4-ylpropylcarbamoyl)methyl]-2,3,4,9-tetrahydro-1H-β-carboline-(3R)-(N- hydroxy)-carboxamide hydrochloride. The compound of formula I is obtained by allowing the amine functional group in the indole ring of the compound of formula II to react with a compound of the formula: Z-(CH2 )m -CO2 Ra (Z is halogen; Ra is a 1-6C alkyl) to form a compound of formula III, deprotecting the amine functional group on its side chain, cyclizing the product to a compound of formula IV, allowing the product of formula IV to condense with the compound of formula V, further allowing the condensation product to condensing with an O-substituted hydroxylamine, then deprotecting the carboxylic functional group, condensation with an amine followed by deprotection of hydroxylamine functional group. |
