Methods have been developed for isolating and proliferating insulin-secreting mammalian cells. Invention methods involve isolating insulin-secreting cells and preparing a primary culture in which insulin is secreted under the control of one or more insulin promoter transcription factors, e.g., NKX 6.1, HNF3-beta, STF-1, Beta 2, and the like. Such proliferated insulin-secreting cells are distinguished from all other cells in that they are proliferated and in that they have measurable intracellular levels of one or more transcription factors associated with insulin production, as can readily be determined by suitable assays.
|
