摘要 PURPOSE: To profitably obtain the L-serine useful as a raw material for medicines, cosmetics, chemicals, etc., by reacting glycine with formaldehyde in the presence of an immobilized enzyme produced by immobilizing a serine hydroxymethyl transferase on an ion exchange resin. CONSTITUTION: The method for producing the highly pure L-serine comprises inoculating Escherichia coli MT-10350, etc., on a culture medium, subjecting the bacterium to a shaking culture at 37 deg.C for 15hr, centrifuging the culture solution, bringing the collected cells into contact with a glycine aqueous solution, shaking the mixture at 40 deg.C for 16hr for the extraction of the enzyme, adding acetone to the extraction solution, centrifugally collecting the produced precipitates, dissolving the collected precipitates in a 50mM phosphate buffer solution, diluting the obtained serine hydroxymethyl transferase solution, bringing the diluted solution into contact with DEAE-agarose having tertiary amino groups on the ion exchange groups to immobilize the enzyme to the ion exchange resin, and subsequently reacting glycine with formaldehyde in the presence of the obtained immobilized enzyme at 50 deg.C for 10min with shaking.
