PURPOSE: To simply and immunologlcally determine free TFPI (human tissue factor coagulation system inhibitor) with good reproducibility by using a monoclonal antibody reating with free TFPI but not reacting with lipoprotein bonding type TFPI. CONSTITUTION: A monoclonal antibody is prepared by immunizing a mouse With human rTFPI and fusing the obtained splenic cell and a mouse myeloma cell and selecting a cell reacting with human rTFPI from the obtained fused cell (hybridoma) to culture the same. By constructing a sandwich immunoassay by utilizing this monoclonal antibody, the simple determination method measuring only free TFPI becomes possible. At this time, so far as an antibody reacting with human TFPI is used as other one kind of an antibody, there is no special limit. As a detection means, enzyme immunoassay, radioimmunoassay, fluorescence immunoassay or emission immunoassay can be selected. |
