<p>PURPOSE:To measure the immunocompetence for diagnosis of immunological diseases or the like in high sensitivity with high reproducibility and with no use of a radioisotope by culturing immunomodulation factor-sensitive culture cells in the presence of the specimen to be tested and determining the potency of this specimen to give proliferation activity to the cultured cells. CONSTITUTION:A specimen to be tested such as human serum is added to a culture medium containing bovine fetal calf serum, 2-mercaptoethanol, gentamicin and interleukin-2, then immunomodulation factor-sensitive culture cells such as mouse-derived CTLL-2 or human-derived OA-1 cells showing interleukin-2 dependent proliferation are added to the resultant culture medium to effect subculture at 3-4 day intervals in a CO2 incubator kept at 37 deg.C. Then, the potency of the specimen to give the culture cells proliferation activity is determined by counting the cell number, incorporation of a radioisotope or enzymatic reactions to measure the immunocompetence of the patient, for example, for diagnosis of immunopathies such as cancer in high sensitivity with good reproducibility.</p> |
